ABSTRACT
The present investigation was carried out with fifteen genotypes of coriander (Coriandrum sativum L.) provided by the Department of Plant Breeding and Genetics, RCA, Udaipur. Purified and isolated DNA was subjected to PCR based marker (RAPD) for assessment of genetic diversity. The quality of DNA was determined by calculating ratio between A260 and A280. The ratio between A260 and A280 was observed 1.50-2.00 which is an indicator of moderately good quality of plant DNA. The concentration of DNA ranged from 1689 ng/µl (Rcr-480) to 3495 ng/µl (Rcr-436). 15 decamer random primers selected, of which 12 primers gave good amplified products with template DNA. They gave 64 scorable fragments. Out of 64 scorable bands, 44 bands were polymorphic. The average percentage of polymorphism was found to be 64.58 per cent. The amplified product size ranged between 100 to 1800 bp. From RAPD profiling, similarity matrix was obtained and jaccard's similarity coefficient ranged from 0.40 to 0.83 and on this basis a dendrogram was constructed with UPGMA method. Dendrogram differentiate 15 Coriander genotypes in nine major clusters. Average effective multiplex ratio (2.79), multiplex ratio (3.89), Rp (6.19), MI (1.95), band informativeness (mIb) (1.47) and PIC (0.70) of the primers were calculated for RAPD marker system. In conclusion, RAPD markers are effective in establishing relationship among the genotypes.
