ABSTRACT
Here we report, characterization of partially purified milk clotting cysteine protease enzyme (MCE) obtained from the latex of Euphorbia nivulia Buch.-Ham. MCE was partially purified by centrifugation and fractional precipitation with acetone. The optimum pH range for milk clotting activity was 6.0-6.5 and it exhibited optimal activity at 500C. The isoelectric point of MCE is 3.4. The clotting activity of the MCE was immensely enhanced by CaCl2. The MCE was activated by thiol compounds and inhibited by common thiol-blocking reagents, particularly mercuric chloride and iodoacetic acid. This indicates that the enzyme belongs to cysteine protease family.
