ABSTRACT
Comparative study of RAPD, ISSR and SSR markers analysis were used to establish genetic identities and evaluate genetic diversity among 17 tobacco cultivars. Among these markers, RAPDs generated 205 amplification products, out of which 146 were polymorphic, ISSRs produced 73 amplification products, out of which 65 were polymorphic and SSR produced 54 amplification products, out of which 51 were polymorphic. SSR fingerprinting detected more polymorphic loci (94.4 %) than the ISSR (89.00 %) and RAPD (89.53 %). Polymorphism information content (PIC) for each of the marker systems (0.71 for RAPD, 0.89 for ISSR and 0.37 for SSR) suggested that all the marker systems were diversely effective in determining polymorphisms. The dendrograms constructed using RAPD, ISSR and SSR marker systems were highly correlated with each other as revealed by high Mantel correlation ranged (r= 0.887 to 0.986). Present study suggests these three marker systems were efficient for genetic diversity studies in tobacco.
