ABSTRACT
A protocol for in vitro cloning of mature plants of potential citrus lime and lemon cultivars has been developed. In vitro shoot multiplication obtained through auxillary bud proliferation from mature nodal explants of 4-5 year old trees of potential lime cultivars viz., NRCC acid lime-8, Shahi sharbati acid lime, local acid lime and two lemon cultivars viz Baramasi lemon and Pant lemon. Various combinations of BAP with NAA were used for optimization of shoot proliferation. Shoots were multiplied by repeated transfer of the initial explants upto three passages. Significantly higher number of shoots obtained in Pant lemon (3.42) followed by NRCC-8 acid lime (3.23), Baramasi lemon (2.91) and Shahi sharbati (2.97). Mean days taken for induction of shoots was shortest in Shahi sharbati acid lime (5.95 days) followed by Pant lemon (6.10 days), Baramasi lemon (6.2 days), NRCC-8 acid lime (6.3 days) & common acid lime (6.3 days). Culture media consisted MS medium supplemented with BAP (1.0 mg L-1), kinetin (0.5mg L-1), IBA (4.0 mg L-1) & NAA (1.0 mg L-1) along with 0.2 gm activated charcoal stimulated multiplication of regenerated plants. The cloned shoots rooted with highest rooting percentage in MS medium with IBA 5.0 mg L-1, NAA 1.0 mg L-1, BAP 1.0mg L-1 + 1 gm Charcoal followed by half strength MS medium with NAA 5.0mg L-1 and IBA 1.0 mg L-1. Maximum rooting of regenerants obtained in Pant lemon followed by NRCC-8 Acid lime, local acid lime, Shahi sarbati acid lime and Baramasi lemon. Plantlets survived acclimatization and hardening exhibited normal growth.
