ABSTRACT
Citrus rootstocks are mostly poly embryonic in nature and success of grafting mainly depends upon the rootstock on which it has been propagated. In Citriculture, Troyer citrange has become one of the prime rootstock used for grafting due to it's genetic dwarfness, suitability for high density planting and CTV tolerance. In India, there is shortage of seed mother blocks of this important rootstock. Hence the present study was undertaken to standardize the in vitro propagation protocol of economically important. Troyer citrange (Chettali) rootstock from CCRI rootstock screening studies. Among the in vitro techniques, regeneration from in vitro seedling epicotyl generally ensures a high proliferation rate, allows production of uniform true to type disease free plants. Callus induction was initiated on solidified MS media supplemented with different concentrations of callus inducing growth regulators individually and in combination, with different explants from four week old in vitro grown seedlings. Observations were taken every three days and the effect of different treatments was quantified on the basis of percentage of explants showing response for callus induction. Optimal callus induction response was obtained on Murashige and Skoog medium (MS), supplemented with NAA 15 mg L-1 and Kinetin 1 mg L-1 from all types of explants, with the epicotyl explant showing the highest response. Maximum shoot regeneration response from epicotyl callus was observed in MS medium supplemented with BAP and NAA combination which was highest in epicotyl explants. Root initiation was observed best on MS medium supplemented with IBA 2 mg L-1.
