ABSTRACT
Randomly amplified polymorphic DNA (RAPD) marker was employed for screening and characterization of root-knot nematode resistance gene in all released tobacco cultivars of Gujarat state. Total 120 RAPD primers were screened, out of which only 15 Primers were selected for present study on the basis of number of amplicon generated by individual primer. Screening of all tobacco cultivars using OPE 17 marker showed 349 bp distinguished specific amplicon only in ABT 10 cultivar. This result indicates that presence of this specific 349 bp gene fragment might impart role in development of resistance in ABT 10 cultivar against root-knot nematode disease. Further this fragment can be used to development of sequence characterized amplified regions marker (SCAR) for rapid, reliable and efficient screening of tobacco cultivars against root-knot diseases resistance, as compared to any other conventional method of screening.
